چکیده

Zimmerer, Robert P. (The Pennsylvania State University, University Park), Robert H. Hamilton, and Christine Pootjes. Isolation and morphology of temperate Agrobacterium tumefaciens bacteriophage. J. Bacteriol. 92:746-750. 1966.-Lysogeny was detected in 14 strains of Agrobacterium tumefaciens among 130 bacterial strains tested with strain B-6 used as the host. Partial lysis was observed with 13 additional bacterial strains. Morphological studies of five strains showed that the phage had similar features. A typical phage (Lv-1) had a polyhedralshaped head, approximately 71 by 63 mmu, and a tail, approximately 211 mmu by 9.5 mmu. The phage nucleic acid was found to be deoxyribonucleic acid. The bacteriophage have been designated L (for lysogenic) followed by the bacterial strain designation in lower case letters.

Isolation of morphological mutants of Agrobacterium tumefaciens.

Morphological mutants were isolated from a wild strain of Agrobacterium tumefaciens at a high frequency by treatment with a nitrosoguanidine. Seventeen of the 20 mutants isolated were temperature-sensitive. At 27 C, the mutant cells were rod-shaped and at 37 C, spherical or branched, whereas the wild-type cells were rod-shaped at both temperatures.

Bacteriophage release in a lysogenic strain of Agrobacterium tumefaciens.

Bacteriophage release in a lysogenic strain of Agrobacterium tumefaciens V-1 is temperature-sensitive. At 25 C and 30 C, phage was released in a ratio of 1 plaque-forming unit per 100 bacteria; at 35 C, although bacterial growth was not inhibited, phage release was suppressed. Phage synthesis was induced by heat shock, 42 C for 30 min, ultraviolet irradiation, and mitomycin C. Induction by ultr...

Isolation of Agrobacterium Tumefaciens Strains from Crown Gall Disease on Imported Roses Plants in Qazvin Province

Cultivar of roses (Rosa spp.) has been grown in greenhouses in Qazvin region of Iran for local markets. Agrobacterium tumefaciens strains were isolated and identified from six different samples of roses plants imported from the Netherland to Iran. During August and September of 2012, nearly 2-5% of rose plants in two different greenhouses in the province of Qazvin were observed with crown gall ...

An enrichment technique for auxotrophs of Agrobacterium tumefaciens using a combination of carbenicillin and lysozyme.

A procedure to enrich for auxotrophic and fermentation mutants of Agrobacterium tumefaciens is described. The method is based on the amplification of the killing power of carbenicillin by the addition of lysozyme. Isolation frequencies of some types of mutants are presented, with and without the application of the proposed procedure. The yield of mutants is usually enhanced a hundredfold per en...

Isolation and Characterization of Lytic Bacteriophages from Wastewater against Listeria monocytogenes

Background and Aims: In the current study, two bacteriophages were isolated from the wastewater sources and characterized by lytic activity against the Listeria monocytogenes (L. monocytogenes) bacterium. Materials and Methods:  50 wastewater samples were collected from the disposal sites. The phages were isolated from the treated samples using the double agar overlay method with L. monocytoge...
